VHH antibodies are derived from camelids ( camelids, llamas, alpacas) and sharks, and contain only heavy chain antibodies, which are the smallest binding-capable antibody fragments available, with a size of only 12-15 kDa. camelids have a distinctive immune system, and in addition to possessing the prototypical antibody, their sera contain an antibody that has lost its light chain, which makes them an ideal animal source to obtain the VHH antibodies.VHH is small in size and have good water solubility, stability, acid resistance, low immunogenicity and unique antigenic epitopes that are rapidly cleared from the bloodstream and penetrate deep into tissues. These unique properties make them widely used in biosensing, affinity capture, molecular imaging, diagnostics and therapeutics of diseases.VHHs are usually derived from immune or natural libraries and displayed and screened by phage or yeast libraries. Among them, phage display technology is most commonly used, and specific VHH antibodies can be produced by appropriate screening strategies. In addition, VHH is easy to produce in E. coli on a large scale and has unique antigen-binding properties compared to other recombinant antibodies.
