Yeast display system background

Yeast display is a eukaryotic protein expression technology that leverages the intracellular protein translocation mechanism (GPI anchor) of the yeast surface display system to immobilize and display exogenous antibodies or protein genes on the yeast surface. The yeast surface display system has a complete protein post-translational modification and secretion mechanism, which is more suitable for the folding, modification and transport of humanized proteins. Therefore, antibodies produced by the yeast display system are closer to human native antibodies in in both conformation and function, and offer promising potential for disease treatment applications. 
 
Yeast display system enables the production of antibodies specific for proteins, peptides, small molecules, and T-cell receptors. These antibodies have the advantages of versatility, quantification and therapeutic efficacy. The application of yeast display technology is even more extensive, which can be used not only for yeast display antibody discovery, but also for protein library screening, protein directed evolution, antigen library construction, high-affinity antibody sorting, antibody affinity maturation, vaccine production, immuno-biocatalysis, and biosensors, etc., which has a broad prospect in the fields of diagnosis, disease treatment and medical research. yeast display library construction Process

Our yeast libraries have large capacity and high diversity, which provide a high foundation for the effective screening of specific antibody sequences. We can construct different forms of yeast antibody libraries (IgG, scFv, VHH, Fab antibody libraries), protein libraries, peptide libraries, cDNA libraries, etc. according to your needs. Additionally, we can also develop antibody libraries of various properties, including immune libraries, natural libraries, synthetic libraries, semi-synthetic libraries, and disease antibody libraries. pYD is our commonly used yeast surface display vector.
 
We are capable of constructing yeast libraries with library capacity up to 10^9, with 100% insertion rate and diversity. Before delivering yeast library, we rigorously validate to ensure the diversity, integrity and functionality of the antibody genes. To ensure that the screened antibodies have very high affinity and specificity, we utilize real-time quantitative evaluation of the antibody library. Fluorescence-activated cell sorting (FACS) quantitatively screen for changes in antibody expression and activity, which utilizes distinct markers on both antibody and antigen to determine binding strength and antibody expression on the surface of yeast cells. We use FACS binding assays to quantitatively screen for antibodies with high binding affinity and selectivity. The display efficiency of our yeast display nanobody library by FACS is more than 95%. Typically, yeast screening involves 3-5 rounds of screening and enrichment, allowing us to obtain high antibody affinity antibodies in a short time frame. 
 
The yeast display system has become a powerful tool in directed protein evolution.
