Product Background

IL2RA (Interleukin-2 Receptor Alpha chain), or CD25, is a component of the interleukin-2 receptor (IL-2R) complex, which includes three subunits: CD25 (α subunit): Confers high-affinity binding when combined with the β (CD122) and γ (CD132) subunits. Alone, CD25 binds IL-2 with low affinity. CD122 (β subunit) and CD132 (γ subunit): Together form an intermediate-affinity receptor. The trimeric complex (αβγ) achieves high-affinity IL-2 binding, essential for robust T cell signaling.

Product Description

Basiliximab (trade name Simulect) is a chimeric mouse-human monoclonal antibody to the α chain (CD25) of the IL-2 receptor of T cells. It is used to prevent rejection in organ transplantation,  especially in kidney transplants.

Mechanism of Action

Basiliximab binds with high-affinity to the alpha-subunit (CD25) of the high-affinity IL-2 receptor. This Inhibitionits IL-2 binding,  which Inhibitionits T-cell activation and prevents the body from mounting an immune response against the foreign kidney.

Metabolism

Most likely removed by opsonization via the reticuloendothelial system when bound to lymphocytes,  or by human antimurine antibody production.
