CD70 (CD70 molecule) is a protein encoding gene that encodes a cytokine, which is a ligand for TNFRSF27/CD27 and belongs to the tumor necrosis factor (TNF) ligand family. Inducing the proliferation of co stimulatory T cells, enhancing the production of cell lytic T cells, and promoting T cell activation, related pathways include MIF mediated glucocorticoid regulation and ERK signaling, CD70-CD27 signaling pathway mediated antigen-specific T cell activation and expansion, thereby providing immune surveillance of B cells.

Product Background

CD70, also known as CD27L, is the sole ligand of CD27 and is a tightly regulated transmembrane glycoprotein expressed on B lymphocytes, T lymphocytes, and APC. CD70 shares structural similarities to other members of the TNF superfamily (TNF α, FasL, RANKL, TRAIL, 4-1 BBL, CD30L, and CD40L). In the normal physiological state, CD70 is expressed only in the thymus and lamina propria.

Product Description

Cusatuzumab is a CD70-directed monoclonal antibody under investigation for the treatment of hematological malignancies.

Mechanism of Action

CD70 is transiently expressed on activated lymphocytes and mature dendritic cells, where it serves as a ligand for the CD27 receptor. The CD70-CD27 interaction results in the downstream activation of the NF-κB pathway responsible for cell proliferation and survival. The overexpression of CD70 can result in the proliferation of malignant cells and has been documented in a variety of solid and hematological tumors - elevated urinary concentrations of soluble CD27 (sCD27), a byproduct resulting from the CD70-CD27 interaction, are correlated with a poorer prognosis in patients with non-Hodgkin lymphoma (NHL), Waldenström macroglobulinemia, and acute lymphoblastic leukemia (ALL).
