Product Background

PDGFRA binds platelet-derived growth factors (PDGF-AA, PDGF-BB, PDGF-CC) to activate signaling pathways (e.g., PI3K/AKT, MAPK, JAK/STAT) that regulate cell growth, differentiation, and angiogenesis.

Product Description

Olaratumab,  sold under the brand name Lartruvo,  is a monoclonal antibody medication developed by Eli Lilly and Company for the treatment of solid tumors. It is directed against the platelet-derived growth factor receptor alpha. It was removed from the United States and European Union markets in 2019,  due to insufficient proof of its medical advantage (see below "Medical uses").

Mechanism of Action

Olaratumab blocks ligand-induced tumor cell proliferation,  and Inhibitionits receptor autophosphorylation and ligand-induced phosphorylation of the downstream signaling molecules protein kinase B (Akt) and mitogen-activated protein kinase. PDGFR signalling is a type of tyrosine kinase-mediated pathway that normally regulates cell growth,  chemotaxis,  and mesenchymal stem cell differentiation. It also promotes internalization of PDGFR thus alters the surface levels of PDGFR.

Metabolism

Mainly degraded nonspecifically by proteolytic enzymes.
