TNFRSF4 (Tumor Necrosis Factor Receptor Superfamily Member 4), also known as OX40 or CD134, is a type I transmembrane protein belonging to the TNF receptor superfamily. Key structural features include: Extracellular Domain: Contains three cysteine-rich domains (CRDs) critical for binding its ligand, OX40L (TNFSF4/CD252). Transmembrane Domain: Anchors the receptor to the cell surface. Cytoplasmic Tail: Lacks intrinsic enzymatic activity but recruits TNF receptor-associated factors (TRAFs), particularly TRAF2, TRAF3, and TRAF5, to activate downstream signaling pathways (e.g., NF-κB, PI3K/Akt).
