The antigen-binding fragment of these molecules is comprised of a single variable domain, termed VHH or more commonly, a nanobody.
Llama immunization represents a cornerstone methodology for generating high-affinity VHH binders. The success of any immunization campaign depends heavily on the quality and conformation of the immunogen. For complex targets like multi-pass membrane proteins (GPCRs, Ion Channels), we leverage our specialized membrane proteomics platform to produce antigens in native-like states using liposomes, nanodiscs, or virus-like particles (VLPs).
Healthy llamas are immunized over a period of 6 to 8 weeks. A series of prime and boost injections are administered in combination with specialized adjuvants to stimulate a robust immune response. Blood samples are drawn periodically to monitor serum titers of heavy-chain-only antibodies compared to conventional antibodies via ELISA.
Once peak immune response is confirmed, peripheral blood mononuclear cells (PBMCs) are isolated. Total RNA is extracted, and cDNA is synthesized.
