OX40L is a hexamer complex, which is composed of a trimer and three OX40 proteins, which affect the genes of T cell division and survival by activating downstream NF-κB, PI3K and AKT signaling pathways, and further promote the transcription of cytokine genes and the expression of cytokine receptors, which is very important for cell survival.
OX 40 and mutual influence between OX 40 L, TNFR related (TRAFs) can affect CD4 + T cell tolerance to the regulation of the OX 40 intracellular domain, TRAF family activate NF- κ B and other signaling pathways, cause the effector T cells and prolong survival time, inhibit the differentiation and activity of regulatory T cells, promote and maintain CD4 + cell memory, promote the generation of memory CD8 + cells, interact with a variety of pro-inflammatory mediators, improve autoimmunity in the body.
OX 40 can increase the survival rate of various types of CD cells and promote the expression of cytokines such as interleukin by enhancing the signaling conduction, which causes the T cell immune response of the tumor cells, and then eliminates the tumor cells.On the contrary, if we block the binding of OX 40 to OX 40 L, we can change the antigen-specific T cell response produced by the body itself and reduce the immune activity of self-free disease. Therefore, OX 40 / OX 40 L inhibitors have therapeutic potential for autoimmune diseases.
