Introduction to CD154/CD40L/TNFSF5

CD154 is a ligand of CD40, also known as CD40L / TNFSF5. It is a member of the tumor necrosis factor (TNF) superfamily. It is also a transmembrane protein of 34-39 kDa, which is mainly expressed in activated T cells, activated B cells, and activated platelets. Peripheral blood monocytes, human vascular endothelial cells, smooth muscle cells, and monocyte macrophages during the inflammatory response also express CD154.
Function of Target CD154/CD40L/TNFSF5
CD154-CD40 has important roles in long-term antigen tolerance and allogeneic transplantation. CD154 can produce costimulatory signals in T cells, and after binding with CD40, it can upregulate the synthesis of IL-4 (interleukin-4) and enhance the immune response. Blockade of CD40 / CD154 results in reduced antigen presentation, lack of an inflammatory environment, partial T cell responses, and T cell tolerance. CD154 and CD40 combine, which can initiate the differentiation, maturation, and apoptotic processes of B cells, with important effects on the function and fate of B cells. CD154 and CD40, which can promote the maturation and antigen presentation ability of APC and increase the expression of costimulatory molecules, activation of CD154 is able to induce APC production IL-12, TNF-α, these factors play important roles in immune regulation and inflammatory response.
Gene Pathway of Target CD154/CD40L/TNFSF5
Upon binding to CD40, CD154 activates various downstream signaling pathways through a variety of different signaling mechanisms. Upon binding to CD40, CD154 degraded I-κB protein by controlling IKK activation, thereby secreting NF-κB dimers. NF-κB subsequently enters the nucleus and begins to regulate the expression of various genes, including inflammation-related and immune-related genes. CD154 is also involved in the activation of the MAPK signaling pathway, which can control the phosphorylation of MAPK kinases such as Erk 1 / 2, p38, and Jnk 1 / 2, and further regulate the processes of cell proliferation, differentiation, and apoptosis.
