2B4, also known as CD244, SLAMF4, is a transmembrane protein, a key cell surface receptor that regulates NK cytolytic activity, signals a member of the lymphocyte activating molecule (SLAM) family, and acts as a receptor in immune regulation.2B4, which is expressed on all NK cells (subsets of T cells, monocytes, and basophils). Unlike other activating and inhibitory receptors, the interaction 2B4 with its ligand CD48 has been shown to mediate both activating and inhibitory functions. CD244 is predominantly present in NK cells, is usually involved in costimulation, and is also associated with depletion or inhibition of the TME.

Function of Target CD244/2B4/SLAMF4

CD244 provides stimulatory or inhibitory signals that regulate various immune responses in NK cells, CD8 + T cells, and other immune cells, such as cytotoxicity, cytokine production, and cell-cell interactions. The cellular and molecular effects of CD244 suggest that it may play a role in the onset and progression of multiple immune-related diseases. In antiviral immunity, the normal function of virus-specific CD8 + T cells and NK cells depends on CD244 expression and the delivery of appropriately stimulatory or inhibitory signals. CD 244 Is located on the surface of various immune cells in the tumor microenvironment, contributes to the regulation of immune function, and plays a crucial role in tumorigenesis and progression.

Gene Pathway of Target CD244/2B4/SLAMF4

CD244 is a member of the SLAM family, which transmits different cellular transduction signals on various immune cells through other pathways, thereby regulating the function of these immune cells and influencing the immune response to various diseases. CD244 binds to its ligand CD48 to form a receptor–ligand complex. The bound CD244 recruits and activates downstream signaling molecules via ITIM and ITAM within its cytoplasm. Specifically, the phosphorylation of ITIM can recruit phosphatases, leading to dephosphorylation and inhibiting the activation of immune cells. However, the phosphorylation of ITAM can recruit and activate other kinases to promote the activation and proliferation of immune cells. Ultimately, signaling by CD244 affects the activation, proliferation, cytotoxicity, and other immune functions of NK and T cells. This signaling process plays a key role in the immune response and immune regulation.
