Introduction To CD27/TNFRSF7

CD27, a member of the TNF receptor superfamily, also known as TNFRSF7, is a transmembrane phosphoglycoprotein expressed on CD4 + and CD8 + T cells and constitutively expressed on naive T cells, memory B cells, NK cells, hematopoietic stem cells (HSC), and progenitor cells. Expression increases upon T cell activation and falls from the cell surface, and upon activation forms soluble CD27.

Function of Target CD27/TNFRSF7

As a member of the TNFR superfamily, CD27 is expressed in most T cells, CD27 signaling is controlled by its ligand CD70, and the CD27 – CD70 interaction provides a potent second signal for the initiation of CD4-dependent T cells and CD8-independent T cells during tumor clearance. The indirect killing mechanism mainly releases perforin and cytokines through the action of transmembrane glycoprotein (CD27) on NK cells and transmembrane glycoprotein (CD70) on NK cells, thus activating cytotoxic T lymphocytes (CTL) to further kill tumor cells. The interaction between CD27 and its ligand CD70 (also known as TNFSF7) provides a critical co-stimulatory signal for T cells. Activation of CD27 can promote T cell activation, proliferation, and differentiation, and subsequently enhance the immune response. CD27 is also involved in regulating the balance of the immune system, maintaining immune homeostasis by affecting the number and function of different T-cell subsets (e.g., effector T cells and regulatory T cells).

Gene Pathway of Target CD27/TNFRSF7

Upon binding of CD27 to its ligand CD70, it triggers a series of signaling events. This binding is specific, and CD70 exists as a trimeric form to bind to the three disulfide bridges of CD27. Upon binding to CD70, CD27 transmits a signal into the cell through the signal transduction domain in its cytoplasmic tail. These signals may include the recruitment and activation of the TRAF (TNF-related factors) family, which in turn trigger the activation of downstream signaling pathways. Activation of the CD27 signaling pathway ultimately leads to the production of a series of downstream effects, including T cell activation, proliferation, differentiation, and the production of cytokines. Together, these effects act on the target cells to generate a functional immune response.
