ICOS-L, also known as B7-H2, CD275, is a costimulatory molecule of the B7 superfamily with about 20% homology with B7-1 and B7-2. ICOS-L is highest in B cells in lymphoid follicles, and the receptor for ICOS-L is expressed in ICOS of T cells. CD275 In normal human lymphoid tissues, cells expressing ICOS are considered as follicular center helper T cells (TFH).
Function of Target CD275/B7-H2/ICOSL
CD275 has a dual role as a co-stimulatory / co-inhibitory immune checkpoint molecule. ICOS-L can undergo co-stimulation by binding with CD28. ICOS may play a role in maintaining durable immune responses and is expressed at particularly high levels in reproductive central T cells or TFH. ICOS-L is constitutively expressed by APC as well as multiple nonhematopoietic stem cells. Earlier studies showed that CD275 can stimulate CD4 + and CD8 + T cell proliferation and IFN-γ production, but more recent work suggests that CD275 can reduce IFN-γ and IL-4 expression by inhibiting NFAT, NF-Bᴋ, and AP-1 pathways, inactivating T cell receptor signaling.CD275 is also involved in regulating the polarization and function of immune cells, such as promoting the activity of Tregs (regulatory T cells) and M2 polarization (anti-inflammatory macrophages) while inhibiting the activity of NK cells and CD8 + T cells, which is conducive to immune evasion and pro-tumorigenic phenotype. CD275 expression is dysregulated in multiple malignancies and is associated with a more aggressive disease course and a poorer outcome. In addition to immune function, CD275 is also expressed on osteoblasts and adipocyte progenitors and is involved in the regulation of skeletal growth and adipocyte differentiation.
Gene Pathway of Target CD275/B7-H2/ICOSL
CD275 transmits signals mainly by interacting with ICOS (an inducible costimulatory molecule). After CD275 binds to ICOS, it may initiate the phosphorylation of Akt (protein kinase B) by activating PI3K (phosphatidylinositol 3-kinase), which then activates the mTOR (mammalian target of rapamycin) signaling pathway to promote the proliferation and survival of T cells. ICOS signaling may also be transmitted through members of the JAK (Janus kinase) and STAT (signal transducers and transcriptional activator) families, affecting the differentiation and function of T cells. ICOS signaling may also involve other signaling pathways, such as the MAPK / ERK signaling pathway, and the activation or inhibition of these pathways can jointly regulate the immune response of T cells.
