Introduction To CD326/EpCAM

Epcam (epithelial cell adhesion molecule, also known as CD326, encoded by tumor-associated calcium signaling 1 gene, belongs to GA733 protein family, is a type I transmembrane glycoprotein and isotype Ca 2 + independent cell adhesion molecule, molecular weight 33 ~ 40 kDa, composed of extracellular domain, single transmembrane domain, intracellular domain, EpEX contains an epidermal growth factor-like repeat domain and a thyroglobulin-like repeat domain, is the structural basis of adhesion between isotype molecules. It is expressed at low levels in the basement membrane of a variety of normal epithelial cells (the highest level in the colon), but it is widely expressed on the cancer cell membrane, especially in squamous carcinoma and adenocarcinoma.

Function of Target CD326/EpCAM

Epcam is involved in cell adhesion processes, but its adhesion is weaker than calcium adhesins. EpCAM is a very classic marker of CSCs (cancer stem cells). Tumor cells expressing EpCAM show greater cancerous ability, and greater stem cell properties. EpCAM is involved in tumor immune regulation, and some scientists have inferred that liver cancer with high EpCAM content can resist NK cell-mediated cytotoxic effects through upregulating carcinoembryonic antigen-related cell adhesion molecule 1. Overexpression of CD326 may lead to uncontrolled cell proliferation and enhanced migratory capacity, thus participating in tumorigenesis and progression.

Gene Pathway of Target CD326/EpCAM

EPCAM interacts with two claudin-7 and cLaudin-1 molecules to regulate the function of tight junctions by preventing their degradation. When the EPCAM gene is knocked down, both claudin-7 and claudin-1 are reduced, tight junctions increase and cells are less susceptible to lysis, demonstrating that EpCAM plays an important role in tumor invasion and dissemination. CD326 may activate the proliferative signaling pathway and promote cell proliferation and division by inducing the expression of genes such as c-myc, cyclins A and E. Abnormalities of this signaling pathway may lead to uncontrolled cell proliferation and tumorigenesis.
