The glucocorticoid-induced TNFR-associated protein, GITR, aka TNFRSF18 or CD357, is a member of the TNFR family and is a type 1 transmembrane protein. GITR is also expressed on natural killer (NK), at low levels on B cells, macrophages, and dendritic cells, and can be upregulated by activation, especially on NK cells. In the activation of CD8 + and CD4 + effector T cells, the expression of GIT R was rapidly increased on T cells and effector T cells, reaching the highest levels on activated T cells.

Function of Target CD357/GITR/TNFRSF18

Both GITR and other molecules of the TNFRSF family, which can act as costimulatory receptors and thus become potential targets for enhanced immunotherapy, involve the activation of GITR signaling mediated by TRAF 2 / 5-NF- κ B, at least partly by upregulating the expression of pro-survival molecules of Bcl-xL, leading to reduced apoptosis of Tregs and promoting Tregs survival. GITR co-stimulation enhances the cytotoxic effects of CD8 + T cells and promotes the survival of bone marrow CD8 + memory T cells. The interaction of GITR and its ligand GITRL (GITR Ligand) can affect the inhibitory activity of Tregs. When GITR is activated, it can attenuate the inhibitory function of Tregs, thereby enhancing the immune response. In contrast, when GITR signaling is blocked, the enhanced inhibitory activity of Tregs contributes to the suppression of excessive immune responses.

Gene Pathway of Target CD357/GITR/TNFRSF18

GITR plays an important role in immune regulation, particularly by regulating the function of Tregs. Tregs are a class of T cells capable of suppressing immune responses and are essential for the maintenance of immune homeostasis and for the prevention of autoimmune diseases. The action of the GITR / GITRL axis on Treg appears to be inhibitory in the short term, while long-term overstimulation in vivo favors the amplification and activation of Treg in mice. The costimulatory effects mediated by GITR ultimately enhanced the proliferative and effector functions of T cells, in part due to the upregulation of IL-2R α, IL-2, and IFN γ. Furthermore, co-stimulation of GITR promotes the production of conventional IL-10 from T cells and favors the differentiation of conventional CD4 + T cells into T-helper2 and Treg cells, and these findings support a balanced role of GITR between T-helper and Treg cells. GITRL / GITR mainly participates in the occurrence and progression of inflammation-mediated diseases through activating effector T lymphocytes and regulating regulatory T lymphocytes (Treg). Intervention in the GITRL / GITR pathway has the effect of clearing virus and parasitic infections and enhancing the effect of anti-tumor treatment.
