Introduction To CD52

Drug targets a biomolecule that can directly bind to drugs and then react. Generally, the target refers to a protein, which is related to the cause of disease. The focus of the target began with focusing on tumors and then began to spread to various fields. Introduction to CD52 protein is a common target in medicine.
CD52, also known as CAMPATH-1 antigen, is a glycoprotein composed of 12 amino acids anchored at the glycosylphosphatidylinositol (glycosylphosphatidylinositol, GPI), with a relative molecular mass of 25-29 KD. CD52 in the cell surface of immune cells (e. g. mature lymphocytes, monocytes, natural killer cells, and dendritic cells) and other tissues and cells (such as the male reproductive tract and mature sperm cells), as a non-regulatory membrane glycoprotein widespread expression, but on hematopoietic progenitor cells, red blood cells, or platelets, did not find its existence.

Function of Target CD52

CD52 can utilize the molecular signaling mechanism of TCR to promote intracellular signaling pathways during T-cell activation. The soluble CD 52 molecule directly binds inhibitory sialic acid-binding immunoglobulin-like lectin 10 (Siglec 10) and also mediates immunomodulation. Further studies showed that CD 52 signaling in T cells is dependent on CD 45 and TCR expression, however, CD52 ligation failed to induce PLC γ 1 activation and calcium signaling in Jurkat T cells, which is opposite to TCR ligation. Thus, CD 52 plays a complex role in T cell activation through multiple pathways. CD 52 can function as a costimulatory molecule to activate both effector cells and regulatory T cells. On the other hand, CD 52 acts as a ligand for Siglec-10 to inhibit T cell activation. Meanwhile, soluble CD52 (sCD 52) is a prognostic impact of the CLL (chronic lymphocytic leukemia) marker.

Gene Pathway of Target CD52

CD 52 signaling in T cells is dependent on CD 45 and TCR expression, however, CD52 ligation failed to induce PLC γ 1 activation and calcium signaling in Jurkat T cells, which is contrary to TCR ligation. The immunosuppressive effects of CD4 + T cells in CD 52 are mainly mediated by soluble CD52 released from phospholipase C. Soluble CD52 molecules are directly bound to the inhibitory receptor Siglec-10 and impair phosphorylation of the TCR-related kinases Lck and Zap 70, and subsequent T cell activation. The inhibitory effect of the soluble CD 52 molecules on T cells is also dependent on the presence of the glycan moiety in the CD52 molecule.
