Introduction To CD70/CD27L

CD70, also known as CD27L, is the sole ligand of CD27 and is a tightly regulated transmembrane glycoprotein expressed on B lymphocytes, T lymphocytes, and APC. CD70 shares structural similarities to other members of the TNF superfamily (TNF α, FasL, RANKL, TRAIL, 4-1 BBL, CD30L, and CD40L). In the normal physiological state, CD70 is expressed only in the thymus and lamina propria.

Function of Target CD70/CD27L

In pro-inflammatory states (infection, malignancy, autoimmune status), CD27-CD70 activity increases lymphocyte proliferation and survival with multiple downstream effects. CD27-CD70 signaling was also shown to promote B cell activation and terminal differentiation into plasma cells, increase cytotoxic CD8 + T cell activity, promote TNF- α production by T cells, and increase NK cell activity through production of IFN- γ and IL-2. The indirect killing mechanism is mainly mediated through the action of CD27 and CD70 on NK cells to release perforin and cytokines on NK cells, thus activating cytotoxic T lymphocytes (CTL) to further kill tumor cells.

Gene Pathway of Target CD70/CD27L

Upon binding of CD27 to its ligand CD70, it triggers a series of signaling events. This binding is specific, and CD70 exists as a trimeric form to bind to the three disulfide bridges of CD27. Upon binding to CD70, CD27 transmits a signal into the cell through the signal transduction domain of its cytoplasmic tail. These signals may include the recruitment and activation of the TRAF (TNF-related factors) family, which in turn trigger the activation of downstream signaling pathways. Activation of TRAFs further triggers activation of downstream signaling pathways, including activation of NF- κ B (nuclear factor κ B) and JNK (c-Jun N-terminal kinase) pathways. Activation of these pathways is crucial for regulating processes such as cell proliferation, differentiation, apoptosis as well as immune response. The CD27 – CD70 interaction provides a potent second signal for the initiation of CD4-dependent and CD8-independent T cells during tumor clearance. The interaction between CD70 and CD27 provides a critical co-stimulatory signal for T cells.
