EGFR (epidermal growth factor receptor) is a transmembrane tyrosine kinase receptor located in the cell membrane, which is an important member of the family of tyrosine kinase receptors, which also includes HER-2 / NEU, HER-3, and HER-4. The EGFR gene is located on the short arm of p12-13 of human chromosome 7, contains 28 exons, and has a molecular weight of 170k Da.EGFR structure consists of three parts: extracellular domain (N terminus), membrane penetrating domain (hydrophobic α helix), and intracellular domain (C terminus).

Function of Target EGFR/HER1/ERBB1

Activation of EGFR can promote the growth and differentiation of many cells, including embryonic development, maintenance and repair of adult tissues, and proliferation and metastasis of cancer cells. EGFR is one of the key proteins involved in the information transmission between parental cancer cells and bystander cells in the tumor microenvironment. EGFR is responsible for the maintenance and function of cancer stem cells (CSCs), including stemness, metabolism, immunomodulatory activity, dormancy, and therapeutic resistance. EGFR regulates these pathways through multiple signaling cascades and often cooperates with other RTKs to exert further control roles.

Gene Pathway of Target EGFR/HER1/ERBB1

The epidermal growth factor receptor (EGFR) is a growth factor receptor that induces cell differentiation and proliferation upon activation by binding one of the ligands. The receptor is located on the cell surface, and the binding of the ligand activates the tyrosine kinase in the intracellular region of the receptor. This tyrosine kinase phosphorylates many intracellular substrates, activating pathways leading to cell growth, DNA synthesis, and expression of oncogenes (such as fos and jun). EGFR activates the JAK / STAT pathway, promotes nuclear transport, stimulates mitosis, and enables the transcription of genes involved in proliferation, leading to cell proliferation and inhibition of apoptosis. Activation of the PI3K / Akt pathway by EGFR mediates cell survival.
