Fab Antibody Library Screening Service

Introduction of Fab Antibody Library Screening ServiceFab

Fab consists of light chain (VL + CL) and heavy chain (VH + CH1), which are connected by interchain disulfide bonds, and consists of 440-450 amino acids with a molecular weight of about 55 kDa. Fab has the advantages of small size, high specificity in tissue distribution, and low immunogenicity, and has been widely used in medical fields such as disease prevention, diagnosis, and treatment. In addition, due to its permeability, Fab antibodies have been widely used in various research fields such as immunohistochemistry, immunofluorescence and flow cytometry, etc. Fab antibodies are usually prepared by using phage display libraries, which can be used to screen Fab antibodies with high affinity and specificity in a short period of time.
Fab antibody-Alpha Lifetech

Alpha Lifetech Can Provide

Alpha Lifetech has many years of experience in phage antibody library construction and screening, and is committed to producing ideal recombinant antibodies for our customers. We have specialized Fab phage screening technology and are happy to provide efficient Fab antibody screening strategies for our customers. We can construct natural and immunized Fab phage antibody libraries for our clients, which are large in capacity and rich in diversity. We clone Fab genes into phage vectors and transform them to obtain Fab phage display libraries. After 2-3 rounds of elution, we can isolate Fab antibodies with strong binding and affinity in about 10 days, which greatly shortens the experimental cycle and facilitates the overall progress of customer projects. We can screen the pre-prepared Fab natural libraries directly without immunization of animals, which is conducive to accelerating the progress of Fab preparation. We can screen a variety of phage libraries based on M13, T4, T7 and λ from human, mouse, rabbit, chicken, sheep and camel species according to the needs of our clients. We generally select Fab antibodies with an affinity of 10^8 after 4 rounds of library screening.

Fab Antibody Library Screening Service Process

Fab antibody-Alpha Lifetech
Alpha Lifetech specializes in the development of phage display and screening technologies, and provides the best phage screening services to our global clients by utilizing comprehensive biological screening strategies and advanced laboratory equipment. We specialize in customized solutions to screen Fab antibodies to the satisfaction of our clients. We specialize in screening large phage Fab antibody libraries. For M13 phage display, we fused Fab fragments with g3p shell proteins on the surface of filamentous phage to screen Fab antibodies with strong affinity for the antigen. We utilized an ELISA-based method to immobilize the antigen, and retained the recombinant phage that bound to the antigen and washed away the weakly binding phage by biological screening. The remaining binding-capable recombinant phages were separated from the antigen by acid treatment and infected with E. coli. E. coli clones expressing phages were infected with helper phages, and Fab phage antibodies capable of binding to the antigen were collected. Antibodies with antigen-binding ability were enriched by repeated phage screening and propagation. During the screening process, phage ELISA is our main assay to confirm positive clones.
