Function of Target Galectin-9/Gal-9/Lgals9
Like other Galectins, Galectin-9 has a variety of biological regulatory functions, including the proliferation, adhesion, aggregation, and apoptosis of tumor cells. By binding to receptors such as Tim-3, Galectin-9 can induce the apoptosis of T cells, thereby inhibiting the activation and proliferation of T cells. Galectin-9 can induce apoptosis in various cell types, including T cells, tumor cells, etc. By promoting apoptosis, Gal-9 plays a role in clearing damaged cells, maintaining tissue homeostasis, and inhibiting tumor growth. The al-9 has adhesive activity and can promote cell – cell interactions or between cells and the extracellular matrix. This adhesive action is important in processes such as embryonic development, tissue repair, and tumor invasion. In the tumor microenvironment, the expression of Gal-9 is often up-regulated, inhibiting the activity of anti-tumor T cells, thus helping tumor cells escape immune surveillance and clearance. Moreover, Gal-9 may also promote tumor progression by promoting angiogenesis and enhancing the migration and invasion capacity of tumor cells.
Gene Pathway of Target Galectin-9/Gal-9/Lgals9
PD-1 interacts with Gal-9 and TIM-3 to attenuate Gal-9 / TIM-3-induced apoptosis of PD-1 + TIM-3 + T cells, Gal-9 is upregulated by inflammatory cytokines IFN β and γ, and targeting Gal-9 is an effective strategy for cancer immunotherapy. After Gal-9 binds to Tim-3, it can induce the apoptosis of T cells, thus inhibiting the immune response of T cells. This signaling pathway plays an important role in the induction of immune tolerance, the prevention and treatment of autoimmune diseases, and tumor immunotherapy. It was found that PD-1, by binding to Gal-9, contributes to the persistence of PD-1 + TIM-3 + T cells and attenuates cell death in Gal-9 / Tim-3-induced cells. This synergy reveals a critical role of PD-1 in the survival of depleted T cells and establishes Gal-9 as an important regulator of the tumor immune response.
