Introduction To Gi24/B7-H5/VISTA

VISTA, where V-domain Ig suppressor of T cell activation is one of the B7 family members, also called B7-H5, B7H5, C10orf54, Days 1, GI24, PD-1H, SISP 1, VSIR, shares 10 – 18% identity and 23 – 33% similarity at the amino acid level with other B7 family members, and phylogenetically forms group B7 family III with B7x and B7H3. As a new member of the immunoglobulin superfamily, the coding gene located on human chromosome 3q13.13, HHLA 2 is a type I transmembrane protein containing 414 amino acids consisting of an extracellular part, a transmembrane region with a tandem IgV 1-IgC-IgV 2 domain, and a cytoplasmic tail. HHLA 2 is expressed in different species, but not in mice and rats, distinguishing it from other members of the B7 family. HHLA 2 protein is usually expressed on antigen-presenting cells (APC), but it has limited expression in normal human organs.

Function of Target Gi24/B7-H5/VISTA

VISTA maintains the quiescence (quiescence) of T cells and myeloid cells in induced immunosuppressive settings in multiple cancer types and is a promising target for combined tumor immunotherapy. As a co-inhibitory ligand, VISTA can directly inhibit T cell function by binding to undefined receptors on T cells. VISTA can also function as a co-inhibitory receptor for T cells to inhibit T cell function independent of antigen-presenting cells (APC).2. Immune-modulatory function. Overexpression of VISTA on bone marrow-derived dendritic cells reduces T cell proliferation and cytokine production, an effect that can be neutralized by VISTA-specific monoclonal antibodies. VISTA plays an important role in T cell co-suppression, immunomodulation, and anti-tumor immune regulation.

Gene Pathway of Target Gi24/B7-H5/VISTA

VISTA signaling on bone marrow cells suppresses myeloid differentiation primary response 88 (MyD 88) -dependent toll-like receptor (TLR) signaling and cytokine production, thereby inducing a more immunosuppressive phenotype. VISTA activity decreases pro-inflammatory cytokines and increases anti-inflammatory cytokines and mediators. The interaction of V-set and Ig domain 3 (VSIG 3) with VISTA on T cells inhibits the activation and proliferation of T cells. VISTA expressed on cancer cells, MDSCs, or tumor-associated macrophages (tam) can bind to PSGL-1 on T cells at acidic pH, but not at neutral pH. PSGL-1 is linked to anti-PSGL-1 (clone 4RA10) to promote T cell exhaustion by reducing extracellular signal-regulated kinase phosphorylation, AKT and transcriptional signaling and cytokine production, and inhibiting T cell receptor (TCR) signaling and activator 5.
