Introduction To HAVCR2/TIM3/TIMD3

Drug targets a biomolecule that can directly bind to drugs and then react. Generally, the target refers to a protein related to the cause of disease. The focus of the target began with focusing on tumors and then began to spread to various fields. Introduction to HAVCR2/TIM3/TIMD3 protein is a common target in medicine.
TIM 3 (T cell immunoglobulin domain and mucin domain-3) is a receptor protein of the TIM family, also known as CD366, HAVCR2 (hepatitis A virus-cell receptor 2), and is a negative regulator of anti-tumor immunity. TIM-3, an important member of the TIM family, was originally found to be a specific marker of fully secreted IFN- γ CD4 + T helper cells 1 (Th 1) and CD8 + cytotoxic T cells. TIM-3 was then also expressed on the cell surface of regulatory T cells (Regulatory cell, Treg), mononuclear macrophages, and dendritic cells (Dendritic cells, DCs).

Function of Target HAVCR2/TIM3/TIMD3

TIM 3 is a kind of negatively regulated immune checkpoint, and its mechanism of action is to play immunosuppressive effects by inhibiting T helper cells (Th 1 and Th 7), inducing CD8 + T cell depletion, and promoting Treg cells to become immunosuppressive cell groups. TIM-3 is expressed on Th 1 and Tc 1 cells, and produces inhibitory signals, thus inhibiting the activation and proliferation of T cells, reducing the production of cytokines that positively regulate the immune response, play a negative regulatory role in the immune process of anti-tumor, and making tumor cells undergo immune escape, which is conducive to tumor invasion into the deep tissue in the original site. TIM-3 is also expressed on certain dendritic cells and can mediate phagocytosis and cross-presentation of antigens in apoptotic cells. Tim-3 induces apoptosis in both Thl and cytotoxic T cells and regulates the functions of NK cells, NKT cells, and macrophages. TIM-3 helps to control the excessive activation of the immune response, thereby preventing autoimmune diseases caused by the excessive immune response. It maintains the balance of the immune system by suppressing hyperactivated T cells.

Gene Pathway of Target HAVCR2/TIM3/TIMD3

The Tim-3 / Gal-9 pathway also inhibits macrophage function and downregulates the release of inflammatory factors. Extracellular Gal-9 and Tim-3 interaction on macrophages. Activation of the PI 3 K / Akt pathway after Tim-3 recruitment inhibited the NF-kB and MAPK signaling cascades in Th 1 cells and macrophages, resulting in reduced TNF- α expression. Activation of the Tim-3 / Gal-9 signaling pathway on M1 macrophages leads to the production of proinflammatory cytokines, whereas in M2 macrophages the recruitment of this pathway leads to the induction of anti-inflammatory cytokines.
