Introduction To IL1B

Interleukin 1β (IL-1β) encoded by the IL 1B gene is one of the most prominent cytokines. As an important member of the IL-1 family, the IL1B genome is 7.8 kb, located on the long arm of chromosome 2, and contains 7 exons. It is usually produced by monocytes and macrophages, in addition, almost all nucleated cells, such as B cells, NK cells, T cells cultured in vitro, keratinocytes, dendritic cells, astrocytes, fibroblasts, neutrophils, endothelial cells, and smooth muscle cells can produce IL-1β.

Function of Target IL1B

IL 1B Is an inflammatory cytokine located upstream of the cytokine cascade, is widely believed to be required for efficient initiation of innate immunity and formation of an adaptive immune response to resolve acute inflammation, and is an amplifier of the immune response. It is also a central mediator of cell-cell interactions in the inflammatory tumor microenvironment. IL 1B has the function of regulating immune cells such as dendritic cells, macrophages, and Th 2 cells, and its aggregation in local tissues; promotes the differentiation of Th 17 cells and related factors; promotes eosinophil degranulation, enhances its cytotoxic effects and induced inflammatory reactions. Increase the synthesis, expression, and release of various cytokines and adhesion factors, including IL-18, intercellular adhesion molecules, vascular cell adhesion molecules, E-selectin, etc. An important role in the fibrotic process of local tissues.

Gene Pathway of Target IL1B

The pro-inflammatory cytokine IL 1 B inhibits gastrin expression by activating Smad 7 and subsequently suppresses the nuclear localization of the Smad 3 / 4 complex. A polymorphic promoter variant of the IL 1 B gene can regulate IL 1 B expression, differentially activating Smad 7 and subsequent suppression of gastrin expression, respectively. After IL 1B binds to receptors on target cells, it activates the IL-1 signaling pathway and myeloid differentiation factor-dependent NF- κ B pathway and exerts positive feedback effects to promote transcription of pro-inflammatory cytokines, induce inflammatory response, and cause fever in the body.
