CD25 is the α -chain of the heterotrimeric IL-2 receptor, which is expressed at high levels in many types of hematological malignancies, but at low levels in most solid tumors. CD25 is also highly expressed in activated circulating immune cells and regulatory T cells (Tregs, CD25 molecule is encoded by the gene IL 2 RA, a single-stranded protein containing about 272 amino acid residues, including 21 amino acid residues signal peptide, a mature molecule containing 251 amino acids, multiple cysteines, and 2 N glycosylation sites. The extracellular region of CD25 contains a single Ig-like fold consisting of a type V (variable) region and a C-type (constant) region. This domain is characterized by having a specific binding site for binding to the IL-2 molecule.
Function of Target IL2RA/CD25
Studies in mice lacking IL-2 or its receptors show evidence of T cell immunodeficiency, suggesting that the role of CD25 is to mediate cellular immune responses. Loss of CD25 expression is associated with increased IL-2 production, increased cytotoxic T cell (CTL) function improved Th 1 differentiation, and concomitant upregulation to limit immune responses to autoantigens CD25 is now commonly used as one of several alternative biomarkers to identify regulatory T cells (Tregs) as its expression increases as the number of these inhibitory cytokine-secreting T cells increases. CD25 is one of the earliest T cell markers detectable on thymocytes before CD8 expression. Meanwhile, CD25 has also been proposed as an effector/memory CTL for antigen activation and a potential marker for senescent T cells.

CD25, as a potential therapeutic target, can enhance the immune response by regulating these signaling pathways, especially by affecting the function of regulatory T cells (Tregs). Tregs play an important role in maintaining immune homeostasis, but their hyperactivation may suppress antitumor immune responses in the tumor microenvironment. Therefore, depleting Tregs by targeting CD25 can restore the balance between effector T cells (Teffs) and Tregs, thus improving the efficiency of the antitumor immune response.
