Introduction To IL5 and IL5R

IL-5 is a pleiotropic cytokine, also known as T cell replacement factor, acidophil colony-stimulating factor 1, acidophil differentiation factor, etc. IL5 is mainly secreted and expressed by T2 lymphocytes Th2, innate lymphocytes ILC2, mast cells, and oncocytes. IL-5R consists of the specific IL-5R α subunits and a non-specific β c heterodimer. IL5 is a dimeric protein with a 4-helical motif that acts on target cells by binding a specific IL5 receptor (IL5R), and though IL5 is a homodimer, it can only bind one IL5R receptor molecule, because the IL5R binding site on IL5 extends to intramolecular sides of the dimer, and IL5R three Fn III domains surround the ligand IL-5, like a wrench around a nut, this wrench-like structure increases the interaction surface area between the receptor and the ligand.
Function of Target IL5 and IL5R
IL 5 is mainly secreted by T cells, these cells in inhaled allergens, respiratory viruses airborne pollutants, and other environmental stimuli release IL 5, at the same time IL 5 has a pleiotropic effect on eosinophils, promotes their maturation, activation, survival, migration from the blood and into the respiratory tract, is a powerful pro-inflammatory cytokine. Therefore, IL-5 is the major cytokine affecting the growth, differentiation, recruitment, activation, and survival of eosinophils, and plays a key role in the pathophysiology of eosinophil-enhancing asthma. Its biological effects are mediated through a selective interaction with its receptor, IL-5R.
Gene Pathway of Target IL5 and IL5R
The IL5 receptor is composed of the IL5 receptor α subunit (IL5R α) and the co-receptor β c subunit (the β c subunit is a signal transduction molecule). IL5R α specifically binds IL5, inducing β c recruitment to IL5R, shared with two monomeric cytokine receptors IL3 and granulocyte-macrophage colony-stimulating factor (GM-CSF), and subsequently activates a complex intracellular signaling network composed of JAK 1 / 2-STT 1 / 3 / 5 module, P38 and MAP kinase and NF-ᴋB transcription factors, ultimately leading to eosinophil maturation, survival, and activation.
