Introduction To IL-6 and IL-6R

IL-6, also known as IFN- β2, hepatocyte stimulating factor and hybridoma/plasmacytoma growth factor, is a 25-kDa secreting glycopeptide monocytes and macrophages that are the major producers of IL-6. IL-6 is a 212-amino acid single-stranded protein located on chromosome 7, where IL-6 and IL-6R α bind to form a complex associating with the gp 130 homodimer. The interleukin-6 receptor alpha (IL-6RA) is the α subunit of the IL-6 receptor and is also shared with other interleukin receptors. IL-6RA is a type I transmembrane glycoprotein that forms a complex with the type I transcellular signal transduction protein glycoprotein 130 (CD130) that modulates the biological activity of IL-6 with low affinity.
Function of Target IL-6 and IL-6R
IL-6T is produced by or after stimulation by cells, B cells, hepatocytes, endothelial cells, fibroblasts, keratinocytes, mesangial cells adipocytes, and several tumor cells. IL-6 is a class of cytokines produced by and acting on a variety of cells. Pro-inflammatory and anti-inflammatory cytokines are involved in various biological processes. It is originally produced by leukocytes and functions among leukocytes.IL-6 is a cytokine with pleiotropic activity, which can promote the proliferation and differentiation of a variety of cells and can also accelerate the synthesis of acute phase proteins in hepatocytes. It can also inhibit the growth of the M1 myeloid leukemia cell line, promote its maturation and differentiation, and inhibit the growth of melanoma and breast cancer cells.
Gene Pathway of Target IL-6 and IL-6R
In the classical signaling cascade, only cells expressing the transmembrane 80-kDa α receptor subunit IL-6R are responsive to IL-6. The monomeric complex of IL-6 and mIL-6R binds to the ubiquitously expressed 130-kDa β receptor subunit receptor (gp 130, CD130) and leads to gp 130 dimerization. The soluble form of sIL-6R can be constructed in complex with IL-6 and gp 130. Both ways to produce sIL-6R: sIL-6R can not only bind its ligand IL-6, but also the IL-6 and sIL-6R complex can bind and activate gp 130 on cells that do not carry membrane-bound IL-6R. This process is called IL-6 trans-signaling, which enhances the IL-6 target cell profile and explains the pleiotropic function of IL-6. After DC interaction with the antigen, IL-6, which is already intracellularly bound with IL-6R in DC, is presented to the plasma membrane in trans. Then, the membrane-bound IL-6 / mIL-6R complex on DC interacts with gp 130 on T cells to induce the production of pathogenic Th 17 cells and to inhibit Treg cells via IL-27.
