Introduction To MSLN/Mesothelin

Mesothelin (MSLN) is a glycoprotein proteolytic protein by the Pre-pro-MSLN precursor, containing N-terminal signal peptide, mature megakaryocytic enhancement factor (MPF), furan cleavage sites, mature MSLN and GPI anchor sequences. The extracellular domain of MSLN contains multiple Ig-like domains and GPI anchor domains involved in intercellular interactions and signaling through binding with other proteins. Expression is limited in normal tissues, but it is significantly upregulated in various malignant tumors such as ovarian, breast, colorectal, and pancreatic cancer.
Function of Target MSLN/Mesothelin
Abnormal expression of MSLN plays an active role in malignant transformation and tumor aggressiveness by promoting cancer cell proliferation, local invasion and metastasis, and resistance to cytotoxic-induced apoptosis. Moreover, the high-affinity interaction between MSLN and CA125 leads to heterotypic cell adhesion, which promotes metastasis. MSLN overexpression promotes cell migration and invasion by inducing the activation and expression of the matrix metalloproteinases MMP 7 and MMP 9. There have been reports of MSLN expression in the metastatic site in lung adenocarcinoma patients, and an association with tumor invasiveness and KRAS mutations. These findings strengthen the rationale for targeting cancer cells expressing MSLN with CARS.
Gene Pathway of Target MSLN/Mesothelin
Overexpression of MSLN is sufficient to constitutively activate intracellular pathways where NF- κB, MAPK, and PI3k-kinase promote cell proliferation and resistance to apoptosis. Activation of the MSLN can trigger the activation of the Akt signaling pathway. Activation of Akt inhibits the expression of apoptosis-related proteins (eg, Bad and Bax) and promotes the expression of cell survival-related proteins (e.g., Bcl-xl, and Bcl-2), thus enhancing the survival of cancer cells. MSLN can also regulate the proliferation and migration of cancer cells by activating the ERK 1/2 (extracellular signal-regulated kinase1/2) signaling pathway. MSLN may also affect the biological behavior of cancer cells by activating the JNK (c-Jun N-terminal kinase) signaling pathway.
