Introduction To ROR1

Drug targets a biomolecule that can directly bind to drugs and then react. Generally, the target refers to a protein related to the cause of disease. The focus of the target began with focusing on tumors and then began to spread to various fields. Introduction to  ROR1 is a common target in medicine.
ROR 1, one member of the receptor tyrosine kinase-like orphan receptor (ROR) family, belongs to the type I receptor tyrosine kinase (receptor tyrosine kinase, RTK) family. Unlike the ROR family in nuclear receptors, ROR is a class of membrane receptors. ROR 1 is a transmembrane receptor that contains extracellular parts, transmembrane regions, and intracellular regions. The extracellular part includes the immunoglobulin-like domain (IG), cysteine-rich domain (CRD), and Kringle domain (KD). The intracellular part includes a tyrosine kinase domain, two serine / threonine-rich domains (S / T), and a proline-rich domain (PRD). The proline-rich domain of ROR 1 physically interacts with cohesion proteins by recruiting SH3 domain-containing proteins, including HS1, DOCK 2, and cortactin, responsible for activating cell migration and proliferation signals, causing resistance to apoptosis.

Function of Target ROR1

Upon activation, ROR 1 can phosphorylate target proteins, thereby regulating the activation of cellular signaling pathways and downstream effects. ROR 1 is highly expressed in a variety of tumor cells and promotes tumor cell survival and proliferation through the activation of downstream signaling pathways. ROR 1 can regulate cell division, proliferation, migration, and cell chemotaxis. Meanwhile, ROR 1 is involved in the activation of the NF-KB pathway in tumor cells and promotes cell migration and invasion as well as cancer metastasis. ROR 1 expression can inhibit apoptosis and help tumor cells evade surveillance by the immune system. ROR 1 enhances the metastatic potential of tumors by enhancing the migration and invasion capacity of tumor cells.

Gene Pathway of Target ROR1

ROR 1 can play important roles in a variety of physiological processes by mediating signaling in the non-classical Wnt signaling pathway (non-canonical Wnt pathways), including the regulation of cell division, proliferation, migration, and cell chemotaxis, especially Wnt 5 a. WNT / ROR 1 signaling is induced by binding to a non-canonical WNT ligand that triggers the formation of a complex between ROR 1 and a FZD receptor, respectively. Signal transduction is the phosphorylation of ROR 1 by several kinases, leading to the inhibition of antiapoptotic pathways on one hand and the activation of downstream pathways such as WNT / PCP, MAPK / ERK, PI3K / AKT) or NF- κ B on the other. These either trigger cytoskeletal rearrangements associated with enhanced tumor cell migration or induce a transcriptional response leading to the expression of genes that promote cell proliferation, survival, EMT, or treatment resistance.
