Introduction To Tie-2

Drug targets a biomolecule that can directly bind to drugs and then react. Generally, the target refers to a protein related to the cause of disease. The focus of the target began with focusing on tumors and then began to spread to various fields. Introduction to  Tie-2  is a common target in medicine.
Tie-2 is a tyrosine kinase receptor containing an epidermal growth factor homologous motif, immunoglobulin-like loop, and fibronectin type III repeats, expressed in endothelial and hematopoietic stem cells and bound to the four angiopoietins. The Tie-2 receptor is a transmembrane protein formed by a combination of extracellular, transmembrane, and intracellular regions. The intracellular domain containing the tyrosine kinase domain is a critical part of the Tie-2 receptor for signal transduction. When the ligand binds to Tie-2, it causes conformational changes in the receptor, which in turn activates its tyrosine kinase activity, leading to the activation of downstream signaling pathways.
Tie-2 receptors are widely expressed on the surface of resting endothelial cells, but also by subsets of hematopoietic cells (e.g., tumor-infiltrating myeloid cells), as well as in pericytes, although at lower levels than in endothelial cells. Phosphorylation of Tie-2 is fine-tuned by unique combinations of ligands and other cell surface proteins.

Function of Target Tie-2

The Tie receptor and their angiogenetic ligands act as modulators of angiogenesis and vascular maturation. The Tie-2 signaling pathway is essential for maintaining the proper function and homeostasis of endothelial cells. Tie-2 is involved in regulating the proliferation, migration, and survival of endothelial cells, by participating in the development and progression of several vascular-related diseases. Tie-2 exerts its function through its putative endothelium-specific expression. Unlike Tie-1, the Tie-2 receptor functions as the primary endothelial angiogenin receptor to prevent atherosclerosis.

Gene Pathway of Target Tie-2

Tie signaling controls the link between endothelial cells and pericytes, but the molecular mechanism of action is still unclear. Pericyte Tie-2 controls sprouting angiogenesis in vitro sprouting and in vivo spheroid assays. Tie2 downstream signaling in pericytes involves Calpain, Akt, and FOXO3A. Ng2-Cre-driven deletion of pericyte-expressed Tie2 in mice transiently delays postnatal retinal angiogenesis. Yet, Tie-2 deletion in pericytes results in a pronounced pro-angiogenic effect leading to enhanced tumor growth. It has been found that exosomes derived from high Tie-2 cervical cancer cells induce TEMs through direct transport of Tie-2 and promote tumor angiogenesis.
