GITRL, the GITR ligand, also known as TNFSF18, is a specific binding molecule for the glucocorticoid-induced tumor necrosis factor receptor (GITR), also a member of the tumor necrosis factor receptor superfamily (TNFRSF), and GITRL is a type 2 transmembrane protein. It is generally considered as a trimer, although it can also exist as a monomer or assemble into other multimeric forms. GITRL is mainly expressed by activated antigen-presenting cells, including macrophages, B cells, dendritic cells, and endothelial cells.
Function of Target TNFSF18/GITRL
GITRL is similar to OX 40 L and is very highly expressed mainly on activated antigen-presenting cells (APC) and endothelial cells. When GITRL is bound to GITR, it signals through a complex containing a single TRAF 5 and two TRAF 2, thereby activating the NF- ΚB and MAPK signaling pathways. This co-stimulatory effect mediated by GITR ultimately enhanced the proliferation and effector function of T cells, in part due to the upregulation of IL-2R α, IL-2, and IFN γ. In addition, GITR activation also protects T cells from activation-induced cell death (AICD) and promotes an increase in memory T cells. GITRL / GITR mainly participates in the occurrence and progression of inflammation-mediated diseases through activating effector T lymphocytes and regulating regulatory T lymphocytes (Treg). Intervention in the GITRL / GITR pathway has the effect of clearing virus and parasitic infections and enhancing the effect of anti-tumor treatment.
Gene Pathway of Target TNFSF18/GITRL
As a specific ligand of GITR, GITRL can either directly bind to GITR or form a soluble extracellular segment associated with GITR after digestion. This binding mode allows GITRL to regulate the function of GITR, which then affects the activation and proliferation of immune cells.
The action of the GITR / GITRL axis on Treg appears to be inhibitory in the short term, while long-term overstimulation in vivo favors the amplification and activation of Treg in mice.GITRL / GITR mainly participates in the occurrence and progression of inflammation-mediated diseases through activating effector T lymphocytes and regulating regulatory T lymphocytes (Treg). Intervention in the GITRL / GITR pathway has the effect of clearing virus and parasitic infections and enhancing the effect of anti-tumor treatment.
